Growth Hormone Secretagogues in Perspective

چکیده

Introduction The parenteral administration of human growth hormone (HGH) has become an intervention of great significance in anti-aging medicine. There is now frenetic interest in using reliable G H secretagogues i n the form of peptides, such as GHRP-2. Although age-related sarcopenia is related to HGH deficiency, the use of HGH for the building of "lean body mass" has been questioned, especially in young adults. Growth hormone administration has precipitated ethical concerns and controversies concerning its use in medical practice. There are many Food and Drug Administration (FDA)-approved brands of HGH, and their use is governed by federal law that has been in place for approximately two decades. In brief, criminal use of HGH involves anyone who "knowingly distributes or possesses with the intent to distribute human growth hormone for any use in humans other than the treatment of a disease or other recognized medical condition where such use has been authorized by the Secretary of Health and Human Services (i.e., FDA) and pursuant to the order of a physician" (21 USCs33 [e]).

Novel mechanisms of growth hormone regulation: growth hormone-releasing peptides and ghrelin.

Growth hormone secretion is classically modulated by two hypothalamic hormones, growth hormone-releasing hormone and somatostatin. A third pathway was proposed in the last decade, which involves the growth hormone secretagogues. Ghrelin is a novel acylated peptide which is produced mainly by the stomach. It is also synthesized in the hypothalamus and is present in several other tissues. This en...

Neuroprotective Actions of Ghrelin and Growth Hormone Secretagogues

The brain incorporates and coordinates information based on the hormonal environment, receiving information from peripheral tissues through the circulation. Although it was initially thought that hormones only acted on the hypothalamus to perform endocrine functions, it is now known that they in fact exert diverse actions on many different brain regions including the hypothalamus. Ghrelin is a ...

Comments on “Growth hormone (GH)-releasing hormone and GH secretagogues in normal aging” This issue of Clinical Interventions in Aging contains a quite interesting and informative article about a topic that is popular and also controversial among practitioners of age-management medicine. Appropriate to that controversy, Drs Hersch and Merriam (2008) have asked the question in the title of their...

Growth hormone secretagogues: characterization, efficacy, and minimal bioactive conformation.

Another class of growth hormone (GH) secretagogues has been discovered by altering the backbone structure of a flexible linear GH-releasing peptide (GHRP). In vitro and in vivo characterization confirms these GH secretagogues as the most potent and smallest (M(r) < 500) reported. Anabolic efficacy is demonstrated in rodents with intermittent delivery. A convergent model of the bioactive conform...
